Compound Overview

FBPase-IN-1 is a potent inhibitor of FBPase (fructose-1,6-bisphosphatase) studied for Type 2 diabetes (T2D), with an IC50 of 0.22 μM. It can lower blood glucose levels and improve glucose tolerance, acting by modifying the C128 site and regulating the N125-S124-S123 allosteric pathway of FBPase, thereby affecting its catalytic activity[1]. It is supplied as a white to off-white solid (C6H4N2S4, MW 232.37) at 98.07% purity.

Safety

For Research Use Only. Not for use in diagnostic or therapeutic procedures, and not for human or veterinary use. Handle in accordance with the Safety Data Sheet and your institution's chemical hygiene plan.

In Vitro

SolventSolubilityNotes
DMSO50 mg/mL (215.17 mM)requires sonication; use freshly opened DMSO (absorbed moisture lowers solubility)

Aliquot the stock solution and store it at -80°C (up to 6 months) or -20°C (up to 1 month); stored under nitrogen; avoid repeated freeze-thaw cycles.

In Vivo

Choose the formulation that suits the animal model and route of administration; percentages are volume ratios of the final working solution. Start from a clear DMSO stock (see In Vitro above), add the co-solvents one at a time in the order listed, mixing after each addition, and prepare the working solution fresh on the day of dosing. If precipitation or phase separation occurs, gentle warming or sonication can help.

Protocol 1

Composition10% DMSO + 40% PEG300 + 5% Tween-80 + 45% saline
Result≥ 2.5 mg/mL (10.76 mM); clear solution
How to prepareGives a clear solution at ≥ 2.5 mg/mL (saturation not determined). For 1 mL of working solution: add 100 μL DMSO stock (25.0 mg/mL) to 400 μL PEG300; then 50 μL Tween-80; then 450 μL saline to bring the volume to 1 mL. Saline: dissolve 0.9 g sodium chloride in ddH2O and make up to 100 mL.

Protocol 2

Composition10% DMSO + 90% (20% SBE-β-CD in saline)
Result≥ 2.5 mg/mL (10.76 mM); clear solution
How to prepareGives a clear solution at ≥ 2.5 mg/mL (saturation not determined). For 1 mL of working solution: add 100 μL DMSO stock (25.0 mg/mL) to 900 μL 20% SBE-β-CD in saline. 20% SBE-β-CD in saline: dissolve 2 g SBE-β-CD powder in 10 mL saline until clear (4°C, store up to one week).

Protocol 3

Composition10% DMSO + 90% Corn Oil
Result≥ 2.5 mg/mL (10.76 mM); clear solution
How to prepareGives a clear solution at ≥ 2.5 mg/mL (saturation not determined). Use with caution if continuous dosing will exceed two weeks. For 1 mL of working solution: add 100 μL DMSO stock (25.0 mg/mL) to 900 μL corn oil.

In Vitro

FBPase-IN-1 (1.5625-200 μM, 48 hours) shows low cytotoxicity, with an IC50 of 75.08 μM[1]. Over 0-45 min, FBPase-IN-1 inhibits FBPase in a time-dependent manner, and the IC50 decreases from 0.28 to 0.09 μM, indicating that FBPase-IN-1 is an irreversible covalent inhibitor of FBPase[1].

In Vivo

In 12 h-fasted male ICR mice (n = 6), FBPase-IN-1 (25 mg/kg, IP) shows good FBPase inhibitory activity (IC50 = 0.22 μM) and a satisfying glucose-lowering effect in vivo, with a 54% reduction in blood glucose at 25 mg/kg[1]. In male ICR mice aged 6 or 7 weeks (n = 6), a single IP dose of FBPase-IN-1 at 25 mg/kg for 4 hours lowers blood glucose levels through FBPase targeting and inhibition of the GNG process[1].
