Hieff NGS™ FFPE DNA Repair Reagent is designed to repair the spectrum of chemical damage in Formalin-Fixed Paraffin-Embedded (FFPE) DNA, including deamination, oxidation, and strand breaks. Compatible with Hieff NGS™ Library Preparation Kits (Cat# 12197 / Cat# 12927) and A-tailing module (Cat# 12608), supporting repair of 5 ng to 1 µg of fragmented FFPE DNA. Also compatible with fragmentation enzyme-based library preparation workflows.

FFPE fixation introduces multiple types of DNA damage including cytosine deamination (C→U mutations), oxidative damage (8-oxoguanine), depurination, cross-linking, and strand breaks. This reagent contains an optimized enzyme cocktail targeting the major classes of FFPE-induced damage, restoring DNA to a state compatible with efficient library preparation and accurate sequencing.
5 ng to 1 µg of fragmented FFPE DNA. Lower inputs are supported but may reduce library yield. Input DNA should be quantified by a dsDNA-specific method (e.g., Qubit fluorometer) rather than UV absorbance, as FFPE DNA preparations often contain absorbance-interfering contaminants.
This reagent is validated and specifically optimized for use with Yeasen Hieff NGS™ Library Preparation Kits (Cat# 12197, Cat# 12927) and the A-tailing module (Cat# 12608), as well as fragmentation enzyme-based library preparation workflows. Compatibility with third-party library preparation kits should be independently validated.
The single-buffer system is compatible with both FFPE DNA repair and the subsequent end-repair/dA-tailing module without requiring buffer exchange between steps. This eliminates a cleanup step, reduces total workflow time, minimizes DNA loss between steps, and simplifies protocol execution.
Supported applications include whole-genome sequencing, targeted gene panel sequencing, whole-exome sequencing, and amplicon sequencing from FFPE-derived libraries. The reagent is designed to maximize library complexity and reduce sequence artifacts introduced by unrepaired damage sites, which would otherwise generate false-positive variant calls.
