Fludarabine triphosphate trisodium, also known as F-ara-ATP trisodium, is the active metabolite of Fludarabine and a potent, noncompetitive, specific inhibitor of DNA primase, with an IC50 of 2.3 μM and a Ki of 6.1 μM. It inhibits DNA synthesis by blocking DNA primase and primer RNA formation, and it also inhibits ribonucleotide reductase and DNA polymerase, ultimately leading to cellular apoptosis[1][2]. It is supplied as a colorless to light yellow liquid (C10H12FN5Na3O13P3, MW 591.12) at 99.93% purity.

In Vitro

Fludarabine triphosphate trisodium inhibits polydeoxythymidylate primase activity more potently than the DNA polymerase α/δ activities in the supernatants of CCRF-CEM cells[1]. At 10-50 μM, Fludarabine triphosphate trisodium inhibits incorporation of ATP into primer RNA and of dTTP into DNA to a similar extent[1].
