Compound Overview
Fluorescent DOTAP, also known as NBD-DOTAP, is a cationic lipid that can be used for nucleic acid and protein delivery. It is labeled with the fluorophore NBD (maximum excitation/emission wavelength ~463/536 nm) and can be used in gene delivery systems, drug delivery, cell imaging, and nanocarrier tracking. It is suited for both biological and pharmaceutical formulation research, such as co-delivery of vaccines[1][3]. It is supplied as an orange to reddish brown solid (C36H60ClN5O7, MW 710.34) at 95.95% purity.

Safety
For Research Use Only. Not for use in diagnostic or therapeutic procedures, and not for human or veterinary use. Handle in accordance with your institution's chemical hygiene plan.
In Vitro
| Solvent | Solubility | Notes |
|---|---|---|
| DMSO | 20 mg/mL (28.16 mM) | requires sonication and warming and heat to 60°C; use freshly opened DMSO (absorbed moisture lowers solubility) |
Aliquot the stock solution and store it at -80°C (up to 6 months) or -20°C (up to 1 month); protect from light, stored under nitrogen; avoid repeated freeze-thaw cycles.
In Vivo
Choose the formulation that suits the animal model and route of administration; percentages are volume ratios of the final working solution. Start from a clear DMSO stock (see In Vitro above), add the co-solvents one at a time in the order listed, mixing after each addition, and prepare the working solution fresh on the day of dosing. If precipitation or phase separation occurs, gentle warming or sonication can help.
Protocol 1
| Composition | 10% DMSO + 90% (20% SBE-β-CD in saline) |
|---|---|
| Result | ≥ 2 mg/mL (2.82 mM); clear solution |
| How to prepare | Gives a clear solution at ≥ 2 mg/mL (saturation not determined). For 1 mL of working solution: add 100 μL DMSO stock (20.0 mg/mL) to 900 μL 20% SBE-β-CD in saline. 20% SBE-β-CD in saline: dissolve 2 g SBE-β-CD powder in 10 mL saline until clear (4°C, store up to one week). |

In Vitro
In fusogenic liposomes, hemolysis rises as the concentration of DOTAP increases with Fluorescent DOTAP[2].
In Vivo
In C57BL/6 female mice, Fluorescent DOTAP (0.5% (w/w), s.c., one single dose) raises the number of CD11c+ cells among total lymph node cells by more than 2.5-fold relative to untreated mice at 4 h[3].
| Animal Model | C57BL/6 female mice (6-7 weeks old) with subcutaneous tumors[3] |
|---|---|
| Dosage | 0.5% (w/w) |
| Administration | Subcutaneous injection (s.c.), one single dose |
| Result | Showed neither CD8+ or CD8- cells took up NBD after fluorescent DOTAP injection. Demonstrated the main uptake is by DC (80%) soon after immunization. Induced migration of activated DC to the DLN. Resulted in DC-T cell interactions and eliciting T cell responses. |
