Compound Overview
FtsZ-IN-8 acts as a potent FtsZ inhibitor, promoting FtsZ polymerization while suppressing its GTPase activity, which disrupts bacterial cell division and leads to bacterial cell death. The compound displays bactericidal activity with rapid killing and little apparent tendency to select for bacterial resistance. It also shows low hemolytic activity and low cytotoxicity toward mammalian cells[1]. It has the molecular formula C27H28BrN3O2 and a molecular weight of 506.43 g/mol.

In Vitro
Among the tested Gram-positive bacteria, FtsZ-IN-8 inhibits S. aureus (MRSA), the methicillin-resistant strain (MIC=0.098 μg/mL), B. subtilis (MIC=0.098 μg/mL), and S. pneumoniae (MIC=0.39 μg/mL)[1]. Bacterial growth is suppressed by FtsZ-IN-8 (1-4× MIC; 0-24 h). Additionally, FtsZ-IN-8 (4× MIC; 4 h) disrupts the surface of MRSA ATCC43300 cells, with notable wrinkling and filamentation[1]. FtsZ-IN-8 (4 μg/mL; 10 min; 25 °C) promotes FtsZ polymerization, and at 0.02-0.64 μg/mL (30 min) it inhibits FtsZ GTPase activity in a dose-dependent manner[1]. FtsZ-IN-8 (12.5 μg/mL; 1 h; 37 °C) shows negligible hemolytic activity against human erythrocytes RAW264.7 cells[1].
