The GATA3 Reporter Lentivirus enables stable, quantitative monitoring of GATA3 transcriptional activity through a fluorescent or luminescent readout driven by tandem GATA3 binding elements. Background

GATA3 is a zinc-finger transcription factor of the GATA family that recognizes GATA DNA-binding motifs to control gene expression in development and immunity. It is the master regulator of T-helper 2 (Th2) cell differentiation and is essential for T lymphoid development and immune regulation. GATA3 is also expressed in multipotent hematopoietic stem cells, where it helps balance self-renewal and differentiation, and it has key roles in mammary gland and skin development. GATA family members GATA1, GATA2, and GATA3 share overlapping but distinct binding preferences; GATA1 and GATA2 are central to erythroid and megakaryocytic lineage development and early hematopoietic cell survival. Altered GATA3 function is implicated in immune disorders and breast cancer.

Product Description & Applications

The GATA3 Reporter Lentivirus is a lentiviral transcription-factor reporter system that provides a sensitive fluorescent or luminescent readout of GATA3 transcriptional activity in mammalian cells. The construct contains tandem repeats of consensus GATA3 DNA-binding elements that drive reporter expression in response to GATA3, coupled to a minimal promoter for high signal-to-noise. The particles are purified by PEG precipitation and sucrose gradient centrifugation and efficiently transduce difficult-to-transfect cells, including primary and thawed cultures. A constitutively expressed selection marker (puromycin or blasticidin) supports stable reporter cell line generation, and the choice of reporters supports readout by fluorescence microscopy, flow cytometry, or luminometry for studies of Th2 differentiation, hematopoiesis, immune regulation, and GATA3-targeted compounds.

About This Product

This reporter lentivirus places a d2GFP, EGFP, Firefly Luc, GFP, mCherry, RFP reporter gene under the control of tandem consensus response elements specific for the GATA3 transcription factor, coupled to a minimal TATA-box promoter and a proprietary upstream enhancer that maximizes signal-to-noise. The constitutively expressed selection marker (Puromycin, Blasticidin) and/or secondary reporter enables stable polyclonal cell line generation and flexible readout by fluorescence microscopy, flow cytometry, or luminometry.

Stable integration via the lentiviral backbone ensures consistent, clonally representative reporter expression in dividing and post-mitotic target cells — including primary T cells, macrophages, organoids, and cryopreserved material — eliminating the variability inherent to transient transfection. The self-inactivating LTR design and third-generation packaging minimize insertional mutagenesis risk and ensure biosafety classification at BSL-2.
