Product Overview
This kit can recover high-purity DNA fragments containing no salt or low salt concentration and no impurities such as protein and RNA from various concentrations of agarose gel. The 200 bp–10 kb DNA fragment has a recovery rate of 80% or more, and single-stranded, double-stranded DNA fragments and circular plasmid DNA can be recovered. Recovered DNA can be used directly in restriction digestion, ligation and sequencing reactions.
The excised gel slice is weighed and dissolved in Solution G at 3 µL per mg of gel (300 µL per 100 mg), or 6 µL per mg for gels above 2%, at 55°C for 5–10 min. The solution is loaded onto Adsorption column G, washed with 300 µL of Solution G and twice with 600 µL of Wash Buffer, dried for 2 min at room temperature and eluted with 15–50 µL of buffer, with 30 µL recommended. Recovered DNA has an OD260/OD280 of about 1.8–2.0.

Usage Notes
- Wear gloves when weighing and handling gel slices.
- Ensure the gel is completely dissolved before loading the column.
- Check that the pH of Solution G is 7.5 or below.
- Add ethanol to the Wash Buffer before first use.
- Extend the column drying time when the room temperature is low.
- Do not use polyacrylamide gels.
