DNA Purification Kits

Product Overview

This kit can recover high-purity DNA fragments containing no salt or low salt concentration and no impurities such as protein and RNA from various concentrations of agarose gel. The 200 bp–10 kb DNA fragment has a recovery rate of 80% or more, and single-stranded, double-stranded DNA fragments and circular plasmid DNA can be recovered. Recovered DNA can be used directly in restriction digestion, ligation and sequencing reactions.

The excised gel slice is weighed and dissolved in Solution G at 3 µL per mg of gel (300 µL per 100 mg), or 6 µL per mg for gels above 2%, at 55°C for 5–10 min. The solution is loaded onto Adsorption column G, washed with 300 µL of Solution G and twice with 600 µL of Wash Buffer, dried for 2 min at room temperature and eluted with 15–50 µL of buffer, with 30 µL recommended. Recovered DNA has an OD260/OD280 of about 1.8–2.0.

Storage & Handling

All components are stored at room temperature for 1 year.

Usage Notes

  • Wear gloves when weighing and handling gel slices.
  • Ensure the gel is completely dissolved before loading the column.
  • Check that the pH of Solution G is 7.5 or below.
  • Add ethanol to the Wash Buffer before first use.
  • Extend the column drying time when the room temperature is low.
  • Do not use polyacrylamide gels.

Safety & Regulatory

For research use only. Not intended for clinical, diagnostic or therapeutic use. Follow the manufacturer's manual and standard laboratory safety practice.

Q.How should the Gel DNA Purification Kit be stored and how long is the shelf life?
A.All components are stored at room temperature for 1 year.
Q.What is included in the kit?
A.Each kit supplies Solution G, Wash Buffer, Elution Buffer and Adsorption column G, in 50 T and 200 T sizes. The 50 T kit contains 60 mL of Solution G, 60 mL of Wash Buffer, 5 mL of Elution Buffer and 50 columns.
Q.How much gel can be loaded and what recovery can be expected?
A.A maximum of 300 mg of gel is processed per tube. Fragments of 200 bp–10 kb are recovered at 80% or more, including single-stranded and double-stranded DNA and circular plasmid DNA, with an OD260/OD280 of about 1.8–2.0.
Q.How is the gel slice dissolved?
A.Solution G is added at 3 µL per mg of gel (300 µL per 100 mg), increased to 6 µL per mg for gels above 2%, and the tube is heated at 55°C for 5–10 minutes until the gel is completely dissolved. The pH of Solution G should be 7.5 or below.
Q.Can polyacrylamide gels be used with this kit?
A.No. The manual states that polyacrylamide gels should be avoided; the kit is intended for agarose gels of various concentrations.
Q.Is this product for research use only?
A.Yes. The manual states that the product is for research use only.
