GKT136901 hydrochloride is a potent, selective and orally active inhibitor of NADPH oxidase NOX1/4, with Ki values of 160 nM and 165 nM, respectively. It also acts as a selective, direct scavenger of peroxynitrite and has anti-inflammatory activity, and can be used to study diabetic nephropathy, stroke, and neurodegeneration[1][2][3]. It is supplied as a light yellow to light brown solid (C19H16Cl2N4O2, MW 403.26) at 99.59% purity.

Aliquot the stock solution and store it at -80°C (up to 6 months) or -20°C (up to 1 month); sealed storage, away from moisture; avoid repeated freeze-thaw cycles.

In Vivo
Choose the formulation that suits the animal model and route of administration; percentages are volume ratios of the final working solution. Start from a clear DMSO stock (see In Vitro above), add the co-solvents one at a time in the order listed, mixing after each addition, and prepare the working solution fresh on the day of dosing. If precipitation or phase separation occurs, gentle warming or sonication can help.

In Vitro
In MPT cells, GKT136901 (10 μM; 30 min) significantly attenuates the high-D-glucose-induced increase in O2•− production and H2O2 generation[4]. GKT136901 (10 μM; 30 min) also eliminates the effect of high D-glucose on activation of p38MAP kinase in MPT cells[4]. In HBMECs, GKT136901 (10 μM; 2 h) attenuates oxidative stress induced by methamphetamine (METH)[5]. GKT136901 (10 μM; 2 h) also protects HBMECs from METH-induced blood-brain barrier (BBB) dysfunction[5].
In Vivo
Renoprotective effects of GKT136901 (30-90 mg/kg; daily p.o. for 16 weeks) are seen in a mouse model of Type 2 diabetes[6].
