Enzyme Activity Assay Kit

Enzyme & Substrate

Glucose-6-phosphate dehydrogenase commits glucose to the pentose phosphate pathway, generating the NADPH that sustains reductive biosynthesis and glutathione recycling; its deficiency is the most common human enzymopathy.

Assay Principle

The Glucose-6-Phosphate Dehydrogenase Activity Colorimetric Assay Kit applies a colorimetric format to this measurement, read at OD 460 nm. Hands-on and incubation time is 20-60 minutes.

In the presence of NADP+ or NAD+, it oxidizes glucose-6-phosphate (G6P) to 6-Phosphogluconolactone (6PG) and NADPH or NADH.

This kit reports the activity of the enzyme itself, which is the relevant measure in G6PD-deficiency and pentose-phosphate flux work.

Q.Which reader is required for this activity assay?
A.This kit requires a standard absorbance plate reader. The readout is taken at OD 460 nm.
Q.What does the activity measurement tell me?
A.This kit reports the activity of the enzyme itself, which is the relevant measure in G6PD-deficiency and pentose-phosphate flux work.
Q.Why measure Glucose-6-Phosphate Dehydrogenase Activity?
A.Glucose-6-phosphate dehydrogenase commits glucose to the pentose phosphate pathway, generating the NADPH that sustains reductive biosynthesis and glutathione recycling; its deficiency is the most common human enzymopathy.
Q.Which sample types are validated for this enzyme?
A.The datasheet lists cells, tissues. Other matrices can usually be adapted, but run a spike-recovery check before relying on the values.
Q.How long does one activity run take?
A.Combined incubation and handling time is 20-60 minutes. Keep this interval identical across plates in the same study, since timing differences show up directly in the endpoint.
Q.What is the detection range?
A.The kit is validated for as low as 0.2 mU/mL under datasheet conditions. Dilute samples that read above the range and concentrate those below it rather than extrapolating from the standard curve.
