Guinea Pig Anti-CHRM3 Antibody (#AMR-006-GP) is a highly specific antibody directed against an epitope of the rat protein. The antibody can be used in western blot and immunohistochemistry applications. It has been designed to recognize CHRM3 from mouse, rat and human samples. The antigen used to immunize guinea pigs is the same as Anti-CHRM3 (#AMR-006) raised in rabbit. Our line of guinea pig antibodies enables more flexibility with our products such as multiplex staining studies, immunoprecipitation and more.

Target CHRM3

Overview

Guinea Pig Anti-CHRM3 Antibody is an antibody targeting Cholinergic receptor muscarinic 3, Acetylcholine receptor muscarinic 3, M3 AChR Polyclonal raised in Guinea Pig (Unconjugated). This antibody is commonly used in IHC, WB to detect, localize, or compare expression of the target across samples.

Key elements and design rationale

  • Target: Cholinergic receptor muscarinic 3, Acetylcholine receptor muscarinic 3, M3 AChR (also reported as Cholinergic receptor muscarinic 3, Acetylcholine receptor muscarinic 3, M3 AChR).
  • Immunogen/epitope region: 3rd intracellular loop.
  • Homology note: Human, mouse, bovine, pig - identical (informative for cross-species interpretation).
  • Species reactivity (as provided): Human, Rat, Mouse.
  • Lot quality control (as provided): Western blot analysis.
  • Peptide confirmation: Confirmed by amino acid analysis and mass spectrometry.
  • Blocking peptide: Available for antigen preadsorption control where appropriate.
  • Conjugate/format: Unconjugated (may affect detection channel and background).

These attributes help researchers interpret whether signal reflects the intended target in a given assay and sample context.

Biological background

The action of the neurotransmitter acetylcholine is mediated through two types of receptors, the ionotrophic nicotinic receptors and the metabotrophic muscarinic receptors. The muscarinic receptors belong to the superfamily of 7-TM G-protein-coupled receptors. Five subtypes of muscarinic receptors have been cloned and named m1-m5.1-2The muscarinic receptors are widely distributed throughout the body, but are predominantly expressed within the parasympathetic nervous system and exerts both excitatory and inhibitory control over central and peripheral tissues.1-2 The m3 receptor is expressed in brain, heart, urinary bladder, salivary gland and airway smooth muscle.3Muscarinic receptors participate in a number of physiologic functions such as regulation of heart rate, muscle contraction, cognition, sensory processing and motor control.1 They also participate in learning and memory processing.4-5 The m1 receptors are the most abundant muscarinic subtype in the cortex and striatum.

Research relevance and current trends

  • Mapping receptor/channel localization across neuronal subtypes and subcellular compartments.
  • Linking trafficking or surface expression changes to activity-dependent signaling and plasticity.
  • Using KO/KD or blocking-peptide concepts to strengthen antibody-based target assignment.

Common research applications

  • Western blot (WB): compare target abundance/size across lysates and conditions; consider isoforms/PTMs.
  • Immunohistochemistry (IHC): examine spatial distribution in tissue and relate signal to cell-type composition.

Interpretation typically benefits from comparing matched sample sets (e.g., treated vs control, WT vs KO/KD) and using orthogonal readouts where feasible.

Notes for experimental interpretation

  • Isoforms and post-translational modifications can shift apparent molecular weight or epitope accessibility across samples.
  • Cross-species signal may depend on epitope conservation; consult the provided homology note when selecting models.
  • Permeabilization, fixation, and antigen retrieval can change accessibility of intracellular vs extracellular epitopes.
  • Conceptual control: antigen preadsorption (blocking peptide) can help assess signal dependence on the immunogen region.
  • Provided control suggestions: Negative control: BLP-MR006.
  • Application notes: see product-specific dilution/usage notes and control concepts provided in the dataset.
