Product Overview
H19 dye is a fluorescent dye designed for high-throughput DNA supercoiling and relaxation assays. DNA gyrases convert relaxed circular DNA into supercoiled DNA, and DNA topoisomerase I converts supercoiled DNA into relaxed DNA.
In the presence of magnesium, relaxed DNA suppresses H19 fluorescence much more than supercoiled DNA, so the signal rises when relaxed DNA is supercoiled and falls when supercoiled DNA is relaxed. The difference is used to measure DNA topology changes by topoisomerases and to screen topoisomerase inhibitors in 96-well plates; the assay can be adapted to 384-well plates.
