Product Overview
Heat-Stable Recombinant Human FGF-basic Protein (FGF2, bFGF) is a low-molecular-weight, monomeric FGF2 expressed in human cells. It is designed to give more stable FGF2 signaling during extended cell culture, and is supplied tag-free, animal-free and xeno-free, lyophilized from a 0.2 µm-filtered PBS solution without carrier protein.
Purity is >95% by SDS-PAGE and endotoxin is <0.5 EU/µg. Typical research uses include human iPSC/ESC maintenance and expansion, FGF-responsive progenitor expansion, organoid culture and FGFR signaling studies.
Expression system: Human cells · Heat-stable · Tag-free · Animal-free
Scientific Background
FGF2 binds heparin and acts through fibroblast growth factor receptors (FGFRs), with cell-surface heparan sulfate as a co-receptor. The common low-molecular-weight human form has 154 amino acids and binds several FGFR splice isoforms, including FGFR1c, FGFR2c, FGFR3c and FGFR4. Downstream pathways include RAS–MAPK/ERK, PI3K–AKT and PLCγ.
Native FGF2 is sensitive to prolonged incubation under cell-culture conditions, so stabilized FGF2 variants were developed to keep signaling more constant between medium changes. "Heat-stable" refers to this culture stability; it does not mean the protein is validated for sterilization, heat processing or therapeutic use. Stabilized FGF2 variants are not interchangeable at the same dose, so the concentration should be optimized for each cell line, medium and matrix.
Research Applications
FGF2 is used wherever cells depend on FGF signaling for self-renewal, expansion or survival, and is also a common ligand for FGFR–ERK and PI3K–AKT signaling studies. A heat-stable form may help keep available FGF2 more even between feeds in longer culture workflows.
| Application | Role of FGF2 | Companion signals and conditions | Common readouts |
|---|---|---|---|
| iPSC/ESC maintenance | Self-renewal, proliferation and survival | TGF-β/Activin support, defined matrix and basal medium | OCT4, SOX2, NANOG, morphology, growth rate, karyotype |
| Neural progenitor expansion | Mitogenic and survival signal | EGF and stage-specific pathway modulation | SOX1, PAX6, NESTIN; expansion and differentiation capacity |
| Mesenchymal / stromal culture | Proliferation and phenotype support | Serum-free or defined basal medium; matrix-dependent | Growth kinetics, morphology, identity markers, functional assays |
| Endothelial research | Proliferation and angiogenic signaling | VEGF-165, extracellular matrix, flow or mechanical cues | CD31, VE-cadherin, KDR; migration, tube or sprouting assays |
| Organoid culture | Stage-specific expansion or patterning | EGF, WNT, BMP/Noggin, Activin and tissue-specific signals | Organoid-forming efficiency, size, lineage markers, function |
Planning considerations
- Decide the role of FGF2 in the experiment (maintenance, expansion, survival, patterning or a signaling assay), start from a validated concentration, then run a dose response in the chosen cell line.
- Validate any change in feeding interval in the complete medium. "Heat-stable" alone does not establish a reduced or weekend-free feeding schedule.
- Heparin, heparan sulfate, matrix and other proteoglycans affect FGF2 stability and FGFR signaling, so keep these conditions consistent between experiments.
- When replacing a conventional FGF2, compare equivalent molar concentrations and check both short-term signaling and long-term phenotype.

Biological Activity & Quality Control
Biological activity is measured by the ability of the protein to stimulate proliferation of 3T3 mouse fibroblast cells. Each lot is tested for purity, endotoxin and biological activity before release.
Lot-specific activity and purity data are provided on the Certificate of Analysis (CoA), available from BioHippo on request.
Reconstitution
- Briefly centrifuge the vial before opening.
- Reconstitute in sterile PBS containing 0.1% endotoxin-free recombinant human serum albumin.
- Mix gently; do not vortex aggressively.
- Prepare single-use aliquots under sterile conditions.
- Avoid repeated freeze–thaw cycles.
Storage & Shipping
| Storage | As supplied: −20°C to −80°C for up to 12 months from receipt. After reconstitution: 2°C to 8°C for up to 1 month, or −20°C to −80°C for up to 3 months, under sterile conditions. Avoid repeated freeze–thaw cycles. |
