Enzyme Activity Assay Kit

Enzyme & Substrate

Hexokinase activity is often low in primary tissue and precious samples, where fluorometric detection resolves turnover that colorimetric formats cannot.

Assay Principle

The Hexokinase Activity Fluorometric Assay Kit applies a fluorometric format to this measurement, read at Ex/Em = 503/580 nm. Hands-on and incubation time is 30-60 minutes.

In this assay, HK converts glucose into glucose-6-Phosphate(G6P), which in turn undergoes a series of reactions and induces the Fluorometric-Probe to generate intense fluorescence product at 585 nm.

Fluorometric detection resolves hexokinase turnover in primary tissue where colorimetric signal is marginal.

Q.Which reader is required for this activity assay?
A.This kit requires a fluorescence-capable plate reader. The readout is taken at Ex/Em = 503/580 nm.
Q.What does the activity measurement tell me?
A.Fluorometric detection resolves hexokinase turnover in primary tissue where colorimetric signal is marginal.
Q.Why measure Hexokinase Activity?
A.Hexokinase activity is often low in primary tissue and precious samples, where fluorometric detection resolves turnover that colorimetric formats cannot.
Q.Which sample types are validated for this enzyme?
A.The datasheet lists cells, tissues. Other matrices can usually be adapted, but run a spike-recovery check before relying on the values.
Q.How long does one activity run take?
A.Combined incubation and handling time is 30-60 minutes. Keep this interval identical across plates in the same study, since timing differences show up directly in the endpoint.
Q.What is the detection range?
A.The kit is validated for as low as 0.5 mU/mL under datasheet conditions. Dilute samples that read above the range and concentrate those below it rather than extrapolating from the standard curve.
