Hieff NGS™ OnePot Pro DNA Fragmentation Module provides a new-generation enzymatic fragmentation reagent for Illumina® and MGI® high-throughput sequencing platforms. The single-tube Smearase™ system performs simultaneous DNA fragmentation, end repair, and dA-tailing, eliminating intermediate cleanup steps. Fragmentation products proceed directly to adapter ligation using Hieff NGS™ Fast-Pace DNA Ligation Module (Cat# 12607) or Novel DNA Ligation Module (Cat# 12626). Supports 500 pg–1 µg dsDNA input with minimal GC bias.

Enzymatic fragmentation (Smearase™) performs fragmentation at 37°C without mechanical stress, eliminating the need for specialized sonication equipment such as Bioruptor or Covaris systems. The single-tube format simultaneously performs fragmentation, end repair, and dA-tailing, replacing three separate workflow steps. Enzymatic fragmentation produces fragments with minimal GC bias compared to some mechanical methods.
500 pg to 1 µg of double-stranded DNA from animal, plant, or microbial genomes. For inputs below 50 ng, additional optimization of fragmentation time may be required to achieve the desired fragment size distribution.
Yes. The Smearase™ Fragmentation Module is specifically designed so that fragmentation products can proceed directly to adapter ligation using Hieff NGS™ Fast-Pace DNA Ligation Module (Cat# 12607) or Novel DNA Ligation Module (Cat# 12626) without an intermediate cleanup step. Skipping cleanup reduces total workflow time, minimizes DNA loss, and reduces cost per library.
For FFPE samples, fragmentation time is adjusted based on the DIN (DNA Integrity Number) score or the degree of degradation. Heavily degraded FFPE DNA (low DIN) requires shorter fragmentation times since the DNA is already partially fragmented by the fixation and storage process. For example, a target peak size of 250 bp requires 9–13 minutes for high-quality FFPE samples (DIN > 8.0).
Validated for use with Illumina® (NovaSeq, HiSeq, MiSeq, NextSeq) and MGI® (MGISEQ-2000, DNBSEQ) high-throughput sequencing platforms. Library preparation with this module generates fragments compatible with standard Illumina and MGI adapter sets.
