Hieff NGSTM RNA Cleaner utilizes highly efficient magnetic beads combined with a proprietary buffer system to specifically bind RNA while effectively removing proteins, salts, and other impurities. It is ideal for purifying total RNA samples post-rRNA removal, in vitro transcribed RNA, RNA-labeled products, and synthetic RNAs. The purified RNA is compatible with RNA library preparation, RT-PCR, qRT-PCR, microarray analysis, Northern blotting, and RNA interference experiments.

Four total RNA samples (1.5 μg each) were diluted to 15 μL as unpurified controls. Another 1.5 μg of each sample was purified with 2.2× magnetic beads and eluted in 15 μL DEPC water. Concentrations were measured from 1 μL of each sample. Quality was checked by loading 1.5 μL on a 2100 Bioanalyzer. The 12602 beads showed over 80% recovery efficiency across all samples.

FAQ

Q: What is the loading capacity of this magnetic bead? How much RNA can 1 μL of it bind to?

A: There is no specific reference. You can first refer to the system in the user manual. For a single reaction with a 50 μl volume of sample, using 1.8 to 2 times the magnetic beads (100 μl magnetic beads) can purify 0.5 - 5 μg RNA.

Q: Can this magnetic bead recover small RNA?

A: Yes, small RNA can be recovered. The recovery rate has not been specifically tested. We usually use it for the purification of Total RNA.

Q: How can you distinguish whether the magnetic beads are normal by their appearance?

A: For normal magnetic beads, when they stand still, the microbeads separate from the buffer and gather at the bottom of the bottle. Before using them, shake to mix evenly, and the microbeads are evenly distributed in the buffer. For abnormal magnetic beads, due to factors such as β-mercaptoethanol, DTT, and large pH differences, it affects the uniformity of the magnetic beads, causing the beads to become thick, aggregate into blocks, or have the phenomenon of sticking to the wall.

Q: When the magnetic beads are not opened, they should be stored at 4℃. After opening, they should be stored at room temperature for use. Will this affect the recovery performance of the magnetic beads?

A: Storing at room temperature will have some impact on the recovery performance of the magnetic beads. It is not recommended to leave them at room temperature for too long. After use, it is recommended to store them at 4℃. The separation effect of PEG is easily affected by pH, temperature, etc.

Q: What are the uses of RNA magnetic beads?

A: 12602: RNA purification magnetic beads; 12603: Complete mRNA capture magnetic beads for eukaryotes

Q: What could be the reasons for magnetic bead precipitation?

A: 1) Sample: The sample contains high levels of sugar or protein; the sample buffer and the magnetic bead buffer are incompatible;

2) Magnetic beads: The magnetic bead buffer has been contaminated.

3) Operation: The drying time should not be too long (generally, it should not exceed 5 minutes)

Q: Can the magnetic beads be reused?

A: The product cannot be reused. The ethanol elution step will affect the performance of the surface-active groups on the magnetic beads.

Q: Can DNA purification beads be used instead of RNA purification beads?

A: It is not recommended. DNA magnetic beads for purifying RNA may cause RNA degradation. The system in RNA magnetic beads is more conducive to maintaining the stability of RNA structure and preventing its degradation.

The beads use a proprietary buffer system combined with magnetic bead technology to specifically bind RNA under defined ionic conditions, while proteins, salts, free nucleotides, and other contaminants remain unbound and are removed during wash steps. RNA is eluted with low-salt water or TE buffer.

Generally, RNA Cleaner beads are optimized for inputs typical in rRNA depletion downstream cleanup and in vitro transcription reactions. Store at 2–8°C for up to 12 months. Do not freeze. Allow to equilibrate to room temperature before use. The beads should be gently mixed by inversion or vortexing before each use. Avoid prolonged exposure to temperatures above 25°C.
