Hieff UNICON™ ColorGPS qPCR Master Mix (No ROX) is a 2× real-time quantitative PCR premix featuring high fluorescence intensity, sensitivity, specificity, and amplification yield. Antibody-based Hot Start inhibits non-specific primer annealing during setup. The ColorGPS mixed-dye system monitors pipetting through a color-change mechanism, substantially reducing pipetting errors. Suitable for instruments that do not require ROX passive reference correction.

ColorGPS uses a combination of two dyes with complementary color-change properties. The mixed-dye system changes color upon contact with the aqueous reaction components, providing a visual indicator that the master mix was dispensed into each well. This is distinct from loading dyes — the color change is triggered by the pipetting action itself, allowing real-time verification during plate setup.
Instruments from manufacturers such as Eppendorf, some Qiagen platforms, and certain Bio-Rad models do not require or benefit from ROX passive reference correction. For these instruments, using a mix without ROX avoids introducing an additional fluorescent signal that could interfere with multiplexing or baseline correction algorithms.
This mix demonstrates low SD values between replicate wells, making it suitable for applications requiring precise quantification across replicates, such as reference gene normalization, absolute copy number determination, and clinical assay development.
After the complete reaction system is prepared (master mix + primers + template), the assembled reaction mix remains stable for extended periods before loading into the qPCR instrument, allowing staggered plate preparation in high-throughput settings.
Valid results require: amplification efficiency 90–110%; standard curve R² > 0.98; S-shaped amplification curve with Ct values between 15 and 35; single-peak melting curve; NTC Ct > 35.
