Hieff Unicon Hotstart Direct Taq DNA Polymerase is a hotstart DNA polymerase that is resistant to blood and other inhibitors. The product is blocked by antibodies and has good amplification sensitivity and specificity. After heating at the pre-denaturation temperature, the antibody is completely inactivated and the DNA polymerase activity is released. The amplification induced by nonspecific annealing of primers could be effectively inhibited by using the hotstart Taq enzyme.

a) According to the specific experimental application, it is needed to prepare the corresponding reaction buffer. If a basic buffer (10×) is required, Cat#11374 is recommended.

b) The amount of DNA and the concentration of probes or primers are recommended concentrations. The optimal concentration can be adjusted according to the specific experimental conditions.

c) The amount of enzyme can be adjusted according to the experimental application.

Store this enzyme at -20°C and avoid repeated freeze-thaw cycles to preserve catalytic activity. The product is shipped Dry Ice and remains stable for up to one year from the date of manufacture when stored under recommended conditions. Aliquoting the stock solution into single-use volumes is recommended for enzymes used infrequently to minimize thermal cycling of the bulk stock.

This enzyme is validated for use in PCR & Amplification, Molecular Diagnostics applications. It is suitable for use in both standard laboratory research settings and, where applicable, optimized reaction workflows requiring high specificity, sensitivity, or throughput.

One unit (U) is defined as the amount of enzyme required to incorporate 10 nmol of deoxyribonucleotide in 30 min at 74°C. The Hot Start format employs monoclonal antibody or chemical modification to inhibit polymerase activity below the denaturation temperature.

This enzyme is produced as Recombinant (E. coli) and supplied as a Research Use Only (RUO) reagent. Each lot is subjected to activity assay, purity assessment by SDS-PAGE, and functional validation prior to release. A Certificate of Analysis (CoA) and Safety Data Sheet (SDS) are available on request.

Hot Start formulations use monoclonal antibodies or chemical modification to block polymerase activity at room temperature, preventing non-specific extension during reaction setup. Full activity is restored after initial denaturation (≥95°C). This results in reduced primer-dimer formation, suppression of non-specific amplification products, and improved sensitivity in complex or low-abundance templates.
