Hieff UNICON™Hotstart E-Taq DNA Polymerase (Cat#10726ES) is a hot start DNA polymerase that employs the company's independently developed dual monoclonal antibodies for double closure. This product not only seals the 5'→3' polymerase activity of Taq DNA polymerase but also seals the 5'→3' exonuclease activity. Heating at the pre-denaturation temperature for 30 seconds can completely inactivate the closure antibodies, releasing the DNA polymerase activity and exonuclease activity. The dual closure feature not only effectively prevents non-specific amplification caused by mispairing or primer dimers but also effectively inhibits the decrease in fluorescence signals caused by probe degradation, providing dual protection to make in vitro diagnostic reagents more stable during transportation or use at room temperature. This Taq enzyme supports the development of a primer-probe pre-mixed system and offers higher sensitivity, specificity, and high GC sequence amplification capability compared to traditional qPCR reagents.

Features

High Specificity: Capable of dual encapsulation, it simultaneously inhibits the 5'-3' Taq DNA polymerase activity and the 5'-3' exonuclease activity at room temperature, enhancing the specificity of the reaction.

No Nuclease Contamination: Free from contamination of exonucleases, endonucleases, and RNases.

Suitable for High-Sensitivity PCR Experiments: Exhibits a good linear relationship when the template concentration ranges from 5×10^6 copies/T to 5×10^1 copies/T, with the lowest detection limit reaching 5 copies/T.

Superior Stability: When formulated into a complete premix containing primers and probes, it remains stable after being left at 37°C for 14 days and after 50 freeze-thaw cycles.

Compatible with Rapid Amplification Protocols: 45 cycles, results within 30 minutes.

01 Good Sensitivity and Linear Range

Figure shows that the qPCR master mix prepared with 10726 exhibits good linearity when the template concentration ranges from 5×10^6 copies/T to 5×10^1 copies/TR2=0.9997.

02 Excellent Thermal Accelerated Stability

Figure 3 demonstrates the stability performance of qPCR master mix prepared with 10726ES, using ASFV plasmid DNA as a template under various temperature and time conditions. The results indicate that the qPCR master mix formulated with 10726ES maintains stable performance.

03 Compatible with Rapid Cycling Programs

Figure shows that the qPCR master mix prepared with 10726ES, using ASFV plasmid DNA as a template, can stably amplify and detect the limit concentration of template DNA5 copies/T under both standard80min and fast30min amplification programs.

Store this enzyme at -20°C and avoid repeated freeze-thaw cycles to preserve catalytic activity. The product is shipped Dry Ice and remains stable for up to one year from the date of manufacture when stored under recommended conditions. Aliquoting the stock solution into single-use volumes is recommended for enzymes used infrequently to minimize thermal cycling of the bulk stock.

This enzyme is validated for use in PCR & Amplification, Molecular Diagnostics applications. It is suitable for use in both standard laboratory research settings and, where applicable, optimized reaction workflows requiring high specificity, sensitivity, or throughput.

One unit (U) is defined as the amount of enzyme required to incorporate 10 nmol of deoxyribonucleotide in 30 min at 74°C. The Hot Start format employs monoclonal antibody or chemical modification to inhibit polymerase activity below the denaturation temperature.

This enzyme is produced as Recombinant (E. coli) and supplied as a Research Use Only (RUO) reagent. Each lot is subjected to activity assay, purity assessment by SDS-PAGE, and functional validation prior to release. A Certificate of Analysis (CoA) and Safety Data Sheet (SDS) are available on request.

Hot Start formulations use monoclonal antibodies or chemical modification to block polymerase activity at room temperature, preventing non-specific extension during reaction setup. Full activity is restored after initial denaturation (≥95°C). This results in reduced primer-dimer formation, suppression of non-specific amplification products, and improved sensitivity in complex or low-abundance templates.
