Hieff UNICONTM HotStart Taq DNA Polymerase is a mixture of Hieff UNICONTM Taq Antibody(Catalog #31301ES)and HieffTM Taq DNA Polymerase. Hieff UNICONTM Taq Antibody had a high affinity for HieffTM Taq DNA Polymerase, and the activity of HieffTM Taq DNA Polymerase could still be blocked after being treated at 50℃ for 30 min. The product was completely inactivated by heating at pre denaturation temperature for 30 seconds, releasing DNA polymerase activity. The hot start Taq enzyme can effectively inhibit the amplification caused by non-specific annealing of primers.

Hieff UNICONTM HotStart Taq DNA Polymerase is suitable for hot start PCR and qPCR. HieffTM Taq DNA Polymerase in this product is a heat stable recombinant DNA polymerase with high template affinity, which is very suitable for amplification of low copy template. The amplified product has 3 '- dA and can be easily cloned into T vector.

  • Closed Detection of DNA Polymerase ActivityIn the Hieff UNICONTM HotStart Taq Buffer (Mg2+ Plus) the activity release was less than 5% after incubation at 65℃ for 30 min.
  • DNA Polymerase Activity Release Detection In the HieffUNICONTM HotStart Taq Buffer (Mg2+ Plus) the activity release was higher than 95% when heated at 95℃ for 30 sec.
  • Exonuclease Activity: In 20 μL reaction system, 10 U of this product and 0.6 μg of λ - HindIII were incubated at 74℃ for 1 h, and the electrophoresis band of DNA had no change.
  • Endonuclease Activity: In 20 μL reaction system, 10 U of the product and 0.6 μg of supercoiled pBR322 DNA were incubated at 74 ℃ for 1 h, and the DNA electrophoretic bands did not change.
  • Residual E.coli gDNA Test: The E.coil 16S rDNA gene was amplified by adding 2 U of the product into 50 μL system and using sterile ddH2O as template. After 35 cycles, the amplified products were subjected to 1% agarose gel electrophoresis and EB staining without amplification bands.

This product should be stored at -25~-15℃ for 2 years.

Store this enzyme at -20°C and avoid repeated freeze-thaw cycles to preserve catalytic activity. The product is shipped Dry Ice and remains stable for up to one year from the date of manufacture when stored under recommended conditions. Aliquoting the stock solution into single-use volumes is recommended for enzymes used infrequently to minimize thermal cycling of the bulk stock.

This enzyme is validated for use in PCR & Amplification, Molecular Diagnostics applications. It is suitable for use in both standard laboratory research settings and, where applicable, optimized reaction workflows requiring high specificity, sensitivity, or throughput.

One unit (U) is defined as the amount of enzyme required to incorporate 10 nmol of deoxyribonucleotide in 30 min at 74°C. The Hot Start format employs monoclonal antibody or chemical modification to inhibit polymerase activity below the denaturation temperature.

This enzyme is produced as Recombinant (E. coli) and supplied as a Research Use Only (RUO) reagent. Each lot is subjected to activity assay, purity assessment by SDS-PAGE, and functional validation prior to release. A Certificate of Analysis (CoA) and Safety Data Sheet (SDS) are available on request.

Hot Start formulations use monoclonal antibodies or chemical modification to block polymerase activity at room temperature, preventing non-specific extension during reaction setup. Full activity is restored after initial denaturation (≥95°C). This results in reduced primer-dimer formation, suppression of non-specific amplification products, and improved sensitivity in complex or low-abundance templates.
