Hieff UNICON™ Lyo-Ready I TaqMan Multiplex qPCR Mix (UDG Plus) is a fluorescent quantitative PCR reagent based on a new-generation antibody-based Hot Start Fast Taq DNA Polymerase, designed for lyophilization-compatible IVD formats. Contains lyophilization protectant, enabling direct freeze-drying without reformulation. Integrated UDG (Uracil-DNA Glycosylase) activity prevents aerosol carry-over contamination, critical for clinical diagnostic applications.

Lyophilization (freeze-drying) enables conversion of liquid qPCR reagents into stable, room-temperature-storable dry formats for point-of-care diagnostic cartridges and decentralized testing devices. A lyophilization-ready mix contains specific cryoprotectants that maintain enzyme activity, dNTP integrity, and buffer composition through the freeze-drying and rehydration cycle without performance loss.

Uracil-DNA Glycosylase (UDG) degrades any dU-containing DNA carryover from previous reactions. When dUTP is substituted for dTTP in the PCR reaction, all amplification products contain dU. Before each new run, UDG is active at lower temperatures to digest any carryover amplicon. UDG is inactivated during the initial denaturation step, allowing normal amplification of native (dT-containing) target DNA to proceed. This is a critical contamination control for high-throughput diagnostic laboratories.

Compatible with dual-labeled probes including FAM, HEX, TET, JOE, and ROX fluorophores, as well as Molecular Beacon probes. This broad compatibility allows use across diverse multiplex panel designs with different probe chemistries.

No. This mix does not contain ROX passive reference dye. It is designed for instruments where ROX correction is not required or where the ROX channel is reserved for a probe fluorophore in the multiplex panel.

Lyophilized reagent preparation stability depends on the specific lyophilization protocol and cartridge format, and should be validated by the user for their specific application. As a liquid reagent, this mix is stable at −20°C under standard storage conditions.
