TaqMan probe-based detection provides a second layer of sequence-specific verification beyond primer annealing — the probe must hybridize to its complementary target sequence within the amplicon for signal to be generated. This eliminates false-positive signal from primer dimers and non-specific amplification products, providing higher specificity. Probe-based detection is also essential for multiplex applications using multiple probes with different fluorophores.
UNICON™ Taq DNA Polymerase uses antibody-based Hot Start, blocking polymerase activity at room temperature and releasing full activity upon initial denaturation at ≥95°C. This is particularly important for TaqMan qPCR with low-abundance targets, where any non-specific amplification during setup reduces assay sensitivity.
Valid results require: amplification efficiency 90–110%; R² > 0.98 for standard curves; S-shaped amplification curve with Ct between 15 and 35; NTC Ct > 35. Ct values below 15 may compress the dynamic range.
