This mix supports up to 4-plex detection — up to four different probe-based targets in a single reaction well using spectrally distinct fluorophores (e.g., FAM, HEX, ROX, Cy5). Each probe must have a distinct excitation/emission spectrum detectable by the qPCR instrument fluorescence channels.
This formulation has strong tolerance to common blood-derived inhibitors such as hemoglobin, IgG, and heparin. This allows use of minimally processed clinical samples including extracted DNA from blood, serum, and plasma without additional purification steps beyond standard extraction.
Compatible with standard dual-labeled TaqMan probes (5′ fluorophore + 3′ quencher), including FAM, HEX, ROX, Cy5, and other dual-labeled formats. Molecular Beacon probes are also compatible.
Stable for 2 years at −15 to −25°C. Shipped on dry ice. Freeze-thaw stability validated to 50 cycles. Thermal acceleration stability validated at 37°C for 14 days without significant performance change.
Valid results: amplification efficiency 90–110% per channel; R² > 0.98 per channel; S-shaped amplification curve with Ct 15–35 per channel; NTC Ct > 35 for each target; no cross-channel spectral bleed-through exceeding instrument correction capability.
