Hifair™ III 1st Strand cDNA Synthesis SuperMix for qPCR (gDNA Digester Plus) is a ready-made premix based on Hifair™ III Reverse Transcriptase. The third-generation enzyme offers significantly enhanced thermostability (active up to 60°C), improving reverse transcription efficiency on RNA templates with complex secondary structures. Increased template affinity makes this product effective for low-template-input applications and low-copy target genes.

Contains 5× gDNA Digester Mix for removal of residual genomic DNA from RNA templates, and 4× Hifair™ III SuperMix Plus with all required RT components (buffer, dNTPs, reverse transcriptase).

Hifair™ III has significantly enhanced thermostability (active up to 60°C) compared to Hifair™ II. The higher reaction temperature improves reverse transcription efficiency on RNA templates with complex secondary structures, GC-rich regions, and long mRNAs that form stable hairpin structures resistant to standard RT temperatures.

The 5× gDNA Digester Mix removes residual genomic DNA contamination from RNA preparations before reverse transcription. This eliminates false-positive signal in downstream qPCR caused by amplification of unspliced pre-mRNA or gDNA, which is particularly important for intron-spanning primer designs.

Shipped on dry ice. Store at −15 to −25°C for up to 18 months from the date of manufacture. Avoid repeated freeze-thaw cycles.

Yes. The enhanced template affinity of Hifair™ III makes this kit particularly effective for low-copy gene detection and applications with limited RNA input. The broad linear detection range supports accurate quantification across a wide dynamic range.

This kit uses an engineered M-MLV (Moloney Murine Leukemia Virus) Reverse Transcriptase variant with enhanced thermostability and processivity. Unlike wild-type M-MLV RT, the engineered version retains activity at reaction temperatures up to 60°C.
