Overview

HiSur Carboxyl Beads are hydrophilic magnetic beads with carboxylic acid groups. These surface groups allow covalent amide bond formation to antibodies, proteins, peptides or other biomolecules via primary amine (NH2) groups. Oligonucleotides, antibodies or other ligands with amine groups can be easily coupled to the beads. Activation through carbodimide is required. Attribute to their very large surface area (about three times larger than that of the MonoMag Carboxyl Beads and unique surface coating, HiSur Carboxyl Beads exhibit superior binding capacity and low non-specific binding of protein or nucleic acids.

These’s HiSur Carboxyl Beads (1 µm) are also available in an easy-to-use kit format, allowing researchers to conjugate antibodies, proteins, peptides or other biomolecules of their own choice to these magnetic beads. With the engineered HiSur Carboxyl Beads and the specifically formulated conjugation buffers, the resulting conjugates exhibit superior binding capacity and low non-specific binding. The kit contains sufficient reagents & components for performing at least 5 conjugation reactions. Briefly, the magnetic beads are activated using carbodiimide and N-hydroxysuccinimide followed by conjugation to amino groups that are present on the target protein/ligand. The recommended protocol has been used to successfully conjugate bovine serum albumin, streptavidin or immunoglobulin to these magnetic beads.

HiSur Carboxyl Beads are 1 µm superparamagnetic beads with a high-surface-area polymer coating providing approximately three times the surface area per bead compared to MonoMag. This supports significantly higher carboxyl group density, resulting in greater binding capacity for amine-containing targets per unit volume of beads.

Carboxylic acid groups are activated by EDC (with optional NHS/Sulfo-NHS) to form reactive NHS esters that couple to primary amines (–NH₂) on antibodies, proteins, and oligonucleotides. The high surface density of –COOH groups supports efficient conjugation at lower protein concentrations.

Choose HiSur (1 µm) when you need fast magnetic separation combined with high binding capacity. Choose SuperMag (50–200 nm) when you want smaller particle size with longer suspension time for extended binding kinetics in solution-phase assays.

HiSur Carboxyl Beads are available in 10 mL and 100 mL volumes (10 mg/mL). For larger bulk or OEM quantities, contact support@biohippo.com.
