Compound Overview

HTH-01-091 TFA is a potent and selective inhibitor of maternal embryonic leucine zipper kinase (MELK), with an IC50 of 10.5 nM. It also inhibits PIM1/2/3, RIPK2, DYRK3, smMLCK, and CLK2, and can be used in breast cancer research[1]. It is supplied as a light yellow to yellow solid (C28H29Cl2F3N4O4, MW 613.46) at 98.01% purity.

In Vitro

SolventSolubilityNotes
DMSO100 mg/mL (163.01 mM)requires sonication; use freshly opened DMSO (absorbed moisture lowers solubility)

Aliquot the stock solution and store it at -80°C (up to 6 months) or -20°C (up to 1 month); avoid repeated freeze-thaw cycles.

In Vivo

Choose the formulation that suits the animal model and route of administration; percentages are volume ratios of the final working solution. Start from a clear DMSO stock (see In Vitro above), add the co-solvents one at a time in the order listed, mixing after each addition, and prepare the working solution fresh on the day of dosing. If precipitation or phase separation occurs, gentle warming or sonication can help.

Protocol 1

Composition10% DMSO + 40% PEG300 + 5% Tween-80 + 45% saline
Result≥ 2.5 mg/mL (4.08 mM); clear solution
How to prepareGives a clear solution at ≥ 2.5 mg/mL (saturation not determined). For 1 mL of working solution: add 100 μL DMSO stock (25.0 mg/mL) to 400 μL PEG300; then 50 μL Tween-80; then 450 μL saline to bring the volume to 1 mL. Saline: dissolve 0.9 g sodium chloride in ddH2O and make up to 100 mL.

Protocol 2

Composition10% DMSO + 90% (20% SBE-β-CD in saline)
Result≥ 2.5 mg/mL (4.08 mM); clear solution
How to prepareGives a clear solution at ≥ 2.5 mg/mL (saturation not determined). For 1 mL of working solution: add 100 μL DMSO stock (25.0 mg/mL) to 900 μL 20% SBE-β-CD in saline. 20% SBE-β-CD in saline: dissolve 2 g SBE-β-CD powder in 10 mL saline until clear (4°C, store up to one week).

Protocol 3

Composition10% DMSO + 90% Corn Oil
Result≥ 2.5 mg/mL (4.08 mM); clear solution
How to prepareGives a clear solution at ≥ 2.5 mg/mL (saturation not determined). Use with caution if continuous dosing will exceed two weeks. For 1 mL of working solution: add 100 μL DMSO stock (25.0 mg/mL) to 900 μL corn oil.

In Vitro

Selective inhibition of 4% of the kinases over 90% is seen with HTH-01-091 (1 μM) TFA[1]. HTH-01-091 (0-10 μM, 1 h) TFA is cell permeable and induces MELK degradation[1]. In breast cancer cells, HTH-01-091 (0-10 μM, 3 day) TFA has minor antiproliferative effects[1].

Cell Proliferation Assay[1]

Concentration0, 0.001, 0.01, 0.1, 1.0, and 10 μM
Incubation Time3 day
ResultShowed antiproliferative activities in a panel of breast cancer cell lines, including MDA-MB-468, BT-549, HCC70, ZR-75-1, MCF7, and T-47D cells, with IC50 values of 4.00 μM, 6.16 μM, 8.80 μM, >10 μM, 8.75 μM, and 3.87 μM, respectively.

Western Blot Analysis[1]

Cell LineMDA-MB-468 cells
Concentration0, 0.1, 1.0, and 10 μM
Incubation Time1 h
ResultReduced MELK protein levels in MDA-MB-468 cells; Dose-dependently decreased MELK pull-down by streptavidin beads, demonstrating that the compound is cell permeable and binds to MELK in an ATP-competitive fashion. Had no effect on ERK1/2 pull-down, showing no binding affinity of HTH-01-091 to ERK1/2.
