Scientific Background

This gene encodes one of the three members of the human AKT serine-threonine protein kinase family which are often referred to as protein kinase B alpha, beta, and gamma. These highly similar AKT proteins all have an N-terminal pleckstrin homology domain, a serine/threonine-specific kinase domain and a C-terminal regulatory domain. These proteins are phosphorylated by phosphoinositide 3-kinase (PI3K). AKT/PI3K forms a key component of many signalling pathways that involve the binding of membrane-bound ligands such as receptor tyrosine kinases, G-protein coupled receptors, and integrin-linked kinase.

What expression system was used and how does it affect protein quality?

This protein was produced in Mammalian (HEK293). Mammalian expression enables human-like glycosylation, correct disulfide bond formation, and proper protein folding — all critical for proteins used in binding interaction studies (SPR/BLI) and antibody characterisation. For transmembrane proteins expressed as ectodomains or nanodisc preparations, the expression system directly affects binding conformation and assay performance.

What amino acid region does this construct cover?

The molecular characterisation of this construct is: AKT1(Met1-Ala480) 8×His tag. Cross-reference with the UniProt entry (P31749) to confirm the expressed region includes your domain of interest — receptor-binding domain, extracellular domain, or specific epitope — before ordering.

What is the purity of this protein and how was it verified?

The purity of the protein is greater than 85% as determined by SDS-PAGE and Coomassie blue staining.. A gel image showing the protein band is provided with each lot on the Certificate of Analysis. This high-purity specification is suitable for SPR/BLI kinetics measurements, ELISA standard curve development, and antibody epitope binding studies.

How should I reconstitute and store this protein?

Please refer to the Certificate of Analysis (CoA) for reconstitution instructions for your specific lot. Storage: Store at -20°C to -80°C for 12 months in lyophilized form. After reconstitution, if not intended for use within a month, aliquot and store at -80°C (A Prepare single-use working aliquots after reconstitution to avoid repeated freeze-thaw cycles. Lyophilized protein is stable for 12 months at −80°C from the date of receipt.
