Assay Principle
This sandwich ELISA quantifies human C-C motif chemokine 17 (gene CCL17) in cell culture supernatants, cell lysates, serum and plasma (heparin, EDTA). Strips are pre-coated with a capture antibody. After the sample is added, a biotinylated detection antibody binds the captured protein, and an avidin–biotin–peroxidase complex supplies the enzyme. TMB develops color in proportion to the bound target; the reaction is stopped and read at 450 nm, and concentrations are read from the standard curve.
CCL17 (also called TARC) is a C-C chemokine that binds the receptor CCR4 and preferentially attracts Th2 cells and regulatory T cells. It is produced by dendritic cells and other cells and is elevated in allergic diseases such as atopic dermatitis, where it is commonly measured as a disease activity marker.
