Assay Principle
This sandwich ELISA quantifies human lysosome-associated membrane glycoprotein 1 (gene LAMP1) in cell culture supernatants, serum and plasma (heparin, EDTA). Strips are pre-coated with a capture antibody. After the sample is added, a biotinylated detection antibody binds the captured protein, and an avidin–biotin–peroxidase complex supplies the enzyme. TMB develops color in proportion to the bound target; the reaction is stopped and read at 450 nm, and concentrations are read from the standard curve.
LAMP1 (CD107a) is a heavily glycosylated membrane protein that lines the lysosome and is one of the most widely used lysosomal markers. It supports lysosome biogenesis, acidification and autophagy, in part by restraining the proton channel TMEM175. In NK cells and cytotoxic T cells it appears on the surface after degranulation, so CD107a staining is a standard readout of cytotoxic activity.
