Assay Principle

This sandwich ELISA quantifies human collagen alpha-1(II) chain (gene COL2A1) in cell culture supernatants, serum and plasma (heparin, EDTA). Strips are pre-coated with a capture antibody. After the sample is added, a biotinylated detection antibody binds the captured protein, and an avidin–biotin–peroxidase complex supplies the enzyme. TMB develops color in proportion to the bound target; the reaction is stopped and read at 450 nm, and concentrations are read from the standard curve.

Collagen α1(II) chain (COL2A1) forms type II collagen, the main collagen of hyaline cartilage and the vitreous of the eye. It is essential for skeletal development and for cartilage resistance to compression, and it is a standard chondrocyte marker in cartilage and osteoarthritis research.

Q.Which sample types can be used with this kit?
A.Validated sample types: Cell culture supernatants, serum, plasma (heparin, EDTA). Other sample types should be validated by the user.
Q.What are the sensitivity and assay range?
A.Sensitivity <20 pg/mL; assay range 156.0–10,000 pg/mL. Dilute samples that read above the top standard and re-assay.
Q.How does this assay work?
A.This sandwich ELISA quantifies human collagen alpha-1(II) chain (gene COL2A1) in cell culture supernatants, serum and plasma (heparin, EDTA).
Q.Does the kit cross-react with related proteins?
A.No significant cross-reactivity or interference with COL2A1 analogs in supplier testing; untested analogs cannot be excluded..
