Assay Principle
This sandwich ELISA quantifies human cartilage oligomeric matrix protein (gene COMP) in cell culture supernatants, cell lysates, serum and plasma (heparin, EDTA). Strips are pre-coated with a capture antibody. After the sample is added, a biotinylated detection antibody binds the captured protein, and an avidin–biotin–peroxidase complex supplies the enzyme. TMB develops color in proportion to the bound target; the reaction is stopped and read at 450 nm, and concentrations are read from the standard curve.
Cartilage oligomeric matrix protein (COMP, thrombospondin-5) is a pentameric extracellular matrix glycoprotein abundant in cartilage, tendon and ligament. It binds collagens and fibronectin to stabilize the matrix, mutations cause pseudoachondroplasia, and COMP is often measured as a marker of cartilage turnover in arthritis research.
