Drug Target Assay Kit

DNA polymerase gamma is the main replicative DNA polymerase in human mitochondria. It handles replication, recombination and repair of mitochondrial DNA and has key roles in aging and mitochondrial disease.

Certain antiviral nucleoside analogs are reported to block this enzyme after they are phosphorylated inside cells, which can lead to severe chronic toxicity.

Kit vs Kit Plus

The only difference is the enzyme: DPG100KE includes 33 µl of 100 x human DNA polymerase gamma, while DPG100K has the same reagents without the enzyme. Both are for 100 assays in 384-well format.

Kit (enzyme not included) (DPG100K) 100 assays
Kit Plus (enzyme included) (DPG100KE) 100 assays

Assay Principle

The assay measures the DNA synthesized by DNA polymerase gamma from a DNA template and dNTPs. A fluorescence dye in EDTA is added after the reaction and the signal is read at 535 nm with excitation at 485 nm.

Safety & Handling

For Research Use Only (RUO). Not for use in diagnostic or therapeutic procedures. The Human DNA Polymerase Gamma Assay Kit is shipped frozen. Follow standard laboratory safety practices when handling the reagents. Please contact us to confirm storage conditions.
Q.What does this kit measure?
A.It measures the activity of human DNA polymerase gamma by quantifying the DNA it synthesizes with a fluorescence dye. It supports high throughput screening of polymerase gamma inhibitors, such as testing nucleoside analogs.
Q.What is the difference between the options?
A.The only difference is the enzyme: DPG100KE includes 33 µl of 100 x human DNA polymerase gamma, while DPG100K has the same reagents without the enzyme. Both are for 100 assays in 384-well format.
Q.Is a protocol available?
A.Yes. The manufacturer's protocol (Human DNA Polymerase Gamma Assay, linked under Documents) covers catalog # DPG100K, DPG100KE.
Q.How long does the assay take?
A.About 60 min. See the protocol for the exact steps.
Q.How should the kit be stored?
A.The Human DNA Polymerase Gamma Assay Kit is shipped frozen. Please contact us for long-term storage conditions.
