Overview

Human Gastric Fibroblasts are isolated from stomach tissue of normal, healthy donors. Fibroblasts are responsible for secretion of the extracellular matrix in connective tissue and contribute to ulcer wound healing; gastric fibroblast activity has also been implicated in the development of gastric carcinoma, which makes these cells a practical model for both tissue-repair and tumour-microenvironment work.

Specifications

Species Human (Homo sapiens)
Tissue Stomach (gastric)
Donor Status Normal, healthy donors
Cell Type Fibroblasts
Product Format Cryopreserved
Pack Size 0.5 million cells/vial

Handling

Thawing Immerse the vial in a 37 °C water bath for 1-2 minutes with gentle agitation, transfer into 5 mL growth medium, centrifuge at 1,000 rpm for 5 minutes
Subculture At 80-90% confluence treat with 0.25% Trypsin-EDTA for 3-5 minutes
Reseeding Density 5,000 cells/cm2

Quality Control

  • Identity verified by immunofluorescence staining for fibronectin and vimentin.
  • Tested negative for HIV-1, HBV, HCV, mycoplasma, bacteria, yeast, and fungi.

Common Research Applications

  • Gastric ulcer and wound-healing models.
  • Extracellular matrix deposition and remodelling assays.
  • Tumour-microenvironment co-culture with gastric carcinoma lines.
  • Fibrosis and myofibroblast differentiation studies.

Notes for Experimental Interpretation

  • The vendor limits expansion to no more than 3 passages; plan experiments within that window and record passage number with every readout.
  • Use the recommended growth medium - fibroblast phenotype and matrix output shift with serum and supplement composition.
  • Thaw rapidly and plate immediately; primary fibroblasts lose viability quickly at intermediate temperatures.

Q.What sample source and donor status do these cells come from?
A.They are isolated from human stomach (gastric) tissue of normal, healthy donors, and cryopreserved at passage P1, 0.5 million cells per vial.
Q.How were the cells characterised and what was tested?
A.Identity was verified by immunofluorescence staining for fibronectin and vimentin. The cells tested negative for HIV-1, HBV, HCV, mycoplasma, bacteria, yeast, and fungi.
Q.What medium should I use and how far can I expand them?
A.Use iXCells Fibroblast Growth Medium (MD-0011). Under the iXCells protocol the cells can be expanded for no more than 3 passages.
Q.What is the recommended subculture and reseeding protocol?
A.At 80-90% confluence treat with 0.25% Trypsin-EDTA for 3-5 minutes, then reseed the new vessel at 5,000 cells/cm2.
Q.How do I thaw the vial?
A.Immerse the vial in a 37 °C water bath for 1-2 minutes with gentle agitation, transfer the cells into 5 mL of growth medium, and centrifuge at 1,000 rpm for 5 minutes before plating.
Q.What are these cells typically used for?
A.Gastric ulcer and wound-healing models, extracellular matrix deposition and remodelling assays, tumour-microenvironment co-culture with gastric carcinoma lines, and fibrosis or myofibroblast differentiation studies.
