Assay Principle
This sandwich ELISA quantifies human interleukin-6 receptor subunit beta (gene IL6ST) in cell culture supernatants, serum and plasma (heparin, EDTA, citrate). Strips are pre-coated with a capture antibody. After the sample is added, a biotinylated detection antibody binds the captured protein, and an avidin–biotin–peroxidase complex supplies the enzyme. TMB develops color in proportion to the bound target; the reaction is stopped and read at 450 nm, and concentrations are read from the standard curve.
Glycoprotein 130 (gp130, encoded by IL6ST) is the shared signal-transducing receptor subunit for the IL-6 cytokine family, including IL-6, IL-11, LIF, OSM and CNTF. Upon cytokine binding it forms homodimers or heterodimers with LIFR or OSMR and activates JAK kinases and STAT3 signaling, underlying roles in immunity, hematopoiesis and development.

Sample dilution: start at 1:200. In the supplier’s tests, serum and plasma samples measured about 190–446 ng/mL.
Example standard curve (OD450, pg/mL): 0 = 0.029; 125 = 0.078; 250 = 0.143; 500 = 0.234; 1000 = 0.424; 2000 = 0.846; 4000 = 1.289; 8000 = 1.909. Run a new standard curve on every plate.
Storage & Handling
Store at 4 °C for up to 6 months or −20 °C for up to 12 months. Avoid repeated freeze–thaw cycles. Ships on gel ice and tolerates up to 3 days at room temperature; freeze on arrival.
