Sandwich ELISA · PicoKine®

Assay Principle

This sandwich ELISA quantifies human interleukin-23 subunit alpha (gene IL23A) in cell culture supernatants, serum and plasma (heparin, EDTA). Strips are pre-coated with a capture antibody. After the sample is added, a biotinylated detection antibody binds the captured protein, and an avidin–biotin–peroxidase complex supplies the enzyme. TMB develops color in proportion to the bound target; the reaction is stopped and read at 450 nm, and concentrations are read from the standard curve.

IL-23 p19 (IL23A) is the subunit that combines with p40 (IL12B) to form the cytokine IL-23, released by dendritic cells and macrophages. IL-23 signals through IL-12Rβ1 and IL-23R to activate JAK2, TYK2 and STAT3, supporting the expansion and survival of IL-17–producing Th17 cells. It is a central target in studies of chronic inflammation and autoimmunity.
