Assay Principle
This single-incubation sandwich ELISA quantifies human interleukin-33 (gene IL33) in cell culture supernatants, serum, plasma (heparin, EDTA) and cell lysates. The strips are coated with an anti-tag antibody. Samples and standards are added to the wells together with the kit’s antibody cocktail (Antibody A and Antibody B); the resulting antibody–target complex is held on the plate through the tag. After washing, TMB substrate is added and the HRP label on the detection antibody converts it to a colored product in proportion to the bound target. The reaction is stopped and the plate is read at 450 nm. The supplier markets the Quick format as a 90-minute assay.
Interleukin-33 (IL-33) is an IL-1 family cytokine that acts as an “alarmin” released from damaged epithelial and endothelial cells. Binding to its receptor ST2 (IL1RL1) activates NF-κB and MAPK signaling, promoting type 2 immunity through Th2 cells, mast cells, eosinophils and group 2 innate lymphoid cells. Inside the nucleus, full-length IL-33 also associates with chromatin.
