Assay Principle
This single-incubation sandwich ELISA quantifies human kallistatin (gene SERPINA4) in cell culture supernatants, cell lysates, serum and plasma (heparin, EDTA). The strips are coated with an anti-tag antibody. Samples and standards are added to the wells together with the kit’s antibody cocktail (Antibody A and Antibody B); the resulting antibody–target complex is held on the plate through the tag. After washing, TMB substrate is added and the HRP label on the detection antibody converts it to a colored product in proportion to the bound target. The reaction is stopped and the plate is read at 450 nm. The supplier markets the Quick format as a 90-minute assay.
Kallistatin (SERPINA4) is a plasma serpin that specifically inhibits tissue kallikrein, forming a stable complex with the enzyme. By controlling kallikrein activity it influences kinin generation, and it is studied in vascular biology, inflammation and blood pressure regulation.

Example standard curve (OD450, pg/mL): 0 = 0.010; 156 = 0.132; 312 = 0.225; 625 = 0.431; 1250 = 0.753; 2500 = 1.438; 5000 = 2.208; 10000 = 2.404. Run a new standard curve on every plate.
