Assay Principle
This sandwich ELISA quantifies human transforming growth factor beta-1 proprotein (gene TGFB1) in cell culture supernatants, cell lysates, serum and plasma (heparin, EDTA). Strips are pre-coated with a capture antibody. After the sample is added, a biotinylated detection antibody binds the captured protein, and an avidin–biotin–peroxidase complex supplies the enzyme. TMB develops color in proportion to the bound target; the reaction is stopped and read at 450 nm, and concentrations are read from the standard curve.
TGF-β1 is the prototypical member of the TGF-β family of cytokines. It is secreted in a latent form held by its latency-associated peptide and, once activated, signals through TGFBR1/TGFBR2 and SMAD proteins to control cell growth, differentiation, extracellular matrix production and immune tolerance, making it central to fibrosis, immunology and cancer research.

Sample dilution: start at 1:100. In the supplier’s tests, serum and plasma samples measured about 80 ng/mL.
Example standard curve (OD450, pg/mL): 0 = 0.052; 62.5 = 0.129; 125 = 0.186; 250 = 0.272; 500 = 0.471; 1000 = 0.78; 2000 = 1.245; 4000 = 1.936. Run a new standard curve on every plate.
Storage & Handling
Store at 4 °C for up to 6 months or −20 °C for up to 12 months. Avoid repeated freeze–thaw cycles. Ships on gel ice and tolerates up to 3 days at room temperature; freeze on arrival.

- Microplate Reader capable of reading absorbance at 450nm.

- Pipettes and pipette tips capable of precisely dispensing 0.5 µl through 1 ml volumes of aqueous solutions.

Activating reagent preparation
LAP (TGF-beta1) is mostly contained as inactive form in samples, please activate it before assay. Don’t activate recombinant LAP (TGF-beta1). Solution A: 1N HCI: add 8.33ml of 12N HCI into 91.67ml of H2O. Solution B: 1.2N NaOH/0.5M HEPES: add 12ml of 10N NaOH and 11.9g HEPES into 75ml of H2O, add H2O to adjust volume to 100ml.
