Assay Principle
This sandwich ELISA quantifies human lipoprotein lipase (gene LPL) in cell culture supernatants, serum and plasma (heparin, EDTA). Strips are pre-coated with a capture antibody. After the sample is added, a biotinylated detection antibody binds the captured protein, and an avidin–biotin–peroxidase complex supplies the enzyme. TMB develops color in proportion to the bound target; the reaction is stopped and read at 450 nm, and concentrations are read from the standard curve.
Lipoprotein lipase (LPL) is the main enzyme that breaks down triglycerides in circulating chylomicrons and VLDL. It is made by adipose tissue, heart and skeletal muscle and is carried to the capillary lumen by GPIHBP1, where it releases fatty acids for storage or energy. LPL deficiency causes severe hypertriglyceridemia, and it is central to lipid metabolism research.
