Assay Principle
This sandwich ELISA quantifies human hepatocyte growth factor-like protein (gene MST1) in cell culture supernatants, cell lysates, serum and plasma (heparin, EDTA). Strips are pre-coated with a capture antibody. After the sample is added, a biotinylated detection antibody binds the captured protein, and an avidin–biotin–peroxidase complex supplies the enzyme. TMB develops color in proportion to the bound target; the reaction is stopped and read at 450 nm, and concentrations are read from the standard curve.
Macrophage-stimulating protein (MSP, encoded by MST1, also called hepatocyte growth factor-like protein) is a plasminogen-related growth factor made mainly by the liver and secreted as an inactive precursor. Once cleaved, it binds and activates the receptor tyrosine kinase RON (MST1R) to regulate macrophage activity, cell migration and epithelial responses. It is distinct from the Hippo pathway kinase also called MST1 (STK4).

Sample dilution: start at 1:100. In the supplier’s tests, serum samples measured 107–328 ng/mL.
Example standard curve (OD450, pg/mL): 0 = 0.036; 156 = 0.121; 312 = 0.183; 625 = 0.317; 1250 = 0.543; 2500 = 0.968; 5000 = 1.566; 10000 = 2.037. Run a new standard curve on every plate.
Storage & Handling
Store at 4 °C for up to 6 months or −20 °C for up to 12 months. Avoid repeated freeze–thaw cycles. Ships on gel ice and tolerates up to 3 days at room temperature; freeze on arrival.
