Assay Principle
Tribioscience’s Fast Human Neurofilament-Light Chain (NF-L) ELISA is designed to quantitatively detect human NF-L levels in serum, plasma, and other biological samples. The main feature is that the kit uses our novel proprietary approaches to combine samples and detections into a one-step instead of the complicated traditional methods. It makes the assay simple, easy, accurate, and fast. The Hands-on time can be within 2 hours, with no need for 4-5 hours (Fig. 1). The detection range is from 0.156 to 10 ng/mL. The levels of human NF-L samples are parallel to the standard curves obtained using the kit standards linearly. Therefore, the kit can be used to determine relative mass values for natural human NF-L protein.
Typical Data This standard curve (R 2 =1.000) is provided for demonstration only. A standard curve should be generated for each set of samples assayed. Fig. 3 is an example of typical Data.
Sensitivity The minimum detectable dose (MOD) of human is typically 100 pg/ml. The Intra-assay CV and the Inter-assay CV are ˂10%.
Human Neurofilament-Light Chain (NF-L) ELISA Kit is a Solid-phase ELISA for the quantitative determination of Neurofilament-Light Chain (NF-L). The microplate is pre-coated with a capture antibody specific to the target. Target present in the sample or standard binds the immobilized antibody, is detected with a biotinylated detection antibody and streptavidin-HRP, and quantified colorimetrically against a standard curve.
Kit Components
The kit provides a pre-coated microplate, target standard for standard-curve generation, detection reagents (detection antibody and enzyme conjugate), wash buffer, substrate, and stop solution. Refer to the product datasheet for the complete component list and quantities.

Safety & Handling
This kit is validated for serum, plasma. Other matrices may require additional in-lab validation.
This assay has a sensitivity of 100 pg/mL and a detection range of 0.156–10 ng/mL. Samples reading outside the range should be diluted or concentrated.
The kit provides a pre-coated microplate, target standard for standard-curve generation, detection reagents (detection antibody and enzyme conjugate), wash buffer, substrate, and stop solution. See the datasheet for the full component list and quantities.
A standard microplate reader capable of measuring absorbance at 450 nm is required for this sandwich ELISA.
