Scientific Background
P2RY12 encodes the P2Y purinoceptor 12, a G-protein coupled receptor (GPCR) that primarily responds to adenosine diphosphate (ADP). It couples to Gi proteins, leading to inhibition of adenylyl cyclase and reduction of intracellular cAMP levels. P2RY12 is highly expressed on platelets, where it plays an essential role in platelet activation, aggregation, and thrombus formation. It is the pharmacological target of antiplatelet drugs such as clopidogrel, prasugrel, and ticagrelor. Beyond the circulatory system, P2RY12 is also expressed in microglia within the central nervous system, where it regulates microglial chemotaxis and homeostatic responses.
