Assay Principle

This sandwich ELISA quantifies human cellular tumor antigen p53 (gene TP53) in cell lysates. Strips are pre-coated with a capture antibody. After the sample is added, a biotinylated detection antibody binds the captured protein, and an avidin–biotin–peroxidase complex supplies the enzyme. TMB develops color in proportion to the bound target; the reaction is stopped and read at 450 nm, and concentrations are read from the standard curve.

p53 (TP53) is a tumor suppressor transcription factor that responds to DNA damage and other stresses by activating genes for cell cycle arrest, DNA repair, senescence or apoptosis. It is mutated in about half of human cancers, making it one of the most studied proteins in cancer biology, with rat models widely used alongside human cells.
