Assay Principle

This sandwich ELISA quantifies human platelet-derived growth factor subunit A (gene PDGFA) in cell culture supernatants, cell lysates, serum and plasma (heparin, EDTA). Strips are pre-coated with a capture antibody. After the sample is added, a biotinylated detection antibody binds the captured protein, and an avidin–biotin–peroxidase complex supplies the enzyme. TMB develops color in proportion to the bound target; the reaction is stopped and read at 450 nm, and concentrations are read from the standard curve.

PDGF-A is a polypeptide chain of platelet-derived growth factor that forms PDGF-AA homodimers or PDGF-AB heterodimers with PDGF-B. Acting mainly through PDGFRα, it stimulates proliferation, migration and survival of mesenchymal cells and is needed for development of lung alveoli, the gut, oligodendrocytes and Leydig cells. It is studied in development, wound healing and fibrosis.
