Drug Target Assay Kit

Poly (ADP-ribose) polymerase-1 (PARP-1) responds to DNA damage by making poly ADP-ribose (PAR), which is attached to PARP-1 itself and to many acceptor proteins. Strong PARP-1 activation depletes NAD+ and produces toxic PAR, which can lead to cell death.

PARP-1 is an anticancer drug target and a possible target in other conditions with heavy DNA damage, such as stroke, ischemia and reperfusion.

Kit vs Kit Plus

PAR100KE includes 17 µl of 300 x human PARP-1 and 55 µl of 200 x NAD+; PAR100K contains the assay buffer, DNA and fluorescence dye without the enzyme or NAD+.

Kit (enzyme and NAD+ not included) (PAR100K) 100 assays
Kit Plus (enzyme included) (PAR100KE) 100 assays

Kit Components

Component Kit (enzyme and NAD+ not included) (PAR100K) Kit Plus (enzyme included) (PAR100KE)
10 x Assay buffer 800 µl 800 µl
100 x DNA 55 µl 55 µl
100 x Fluorescence dye 220 µl 220 µl
200 x NAD+ Not included 55 µl
300 x human PARP-1 Not included 17 µl

Assay Principle

The assay measures PAR, the product of PARP-1. PAR binds a fluorescent dye and increases its signal, which is read at 535 nm with excitation at 485 nm.

Safety & Handling

For Research Use Only (RUO). Not for use in diagnostic or therapeutic procedures. The Human Poly (ADP-ribose) Polymerase-1 Assay Kit is shipped frozen. Follow standard laboratory safety practices when handling the reagents. Please contact us to confirm storage conditions.
Q.What does this kit measure?
A.It measures human PARP-1 activity by detecting the poly ADP-ribose (PAR) it makes from NAD+, which raises the signal of a fluorescent dye. It can be used to test PARP-1 inhibitors.
Q.What is the difference between the options?
A.PAR100KE includes 17 µl of 300 x human PARP-1 and 55 µl of 200 x NAD+; PAR100K contains the assay buffer, DNA and fluorescence dye without the enzyme or NAD+.
Q.Is a protocol available?
A.Yes. The manufacturer's protocol (Human poly (ADP-ribose) polymerase-1 assay, linked under Documents) covers catalog # PAR100K, PAR100KE.
Q.How long does the assay take?
A.About 60 min. See the protocol for the exact steps.
Q.How should the kit be stored?
A.The Human Poly (ADP-ribose) Polymerase-1 Assay Kit is shipped frozen. Please contact us for long-term storage conditions.
