Overview

Human Primary Rheumatoid Arthritis Synovial Fibroblasts are human frozen primary cells from connective tissue used for extracellular matrix remodeling, stromal signaling, wound response, and tissue remodeling studies. These cells display adherent , fibroblast growth characteristics. Donor history reported for this listing: Rheumatoid Arthritis.

Key elements and design rationale

  • Biological source: Human (H. sapiens)
  • Tissue origin: Connective Tissue
  • Growth properties: Adherent , fibroblast
  • Format: Frozen
  • Seeding guidance: 7,000 - 10,000 cells/cm²
  • Biosafety level: II
  • Culture context: For optimal cell culture, we recommend using PriCoat™ T25 Flasks (G299) or coating your preferred vessels with Applied Cell Extracellular Matrix (G422) . PriGrow X Series Medium (TM5409) + 1% Penicillin/Streptomycin Solution (G255), 37.0°C, 5% CO₂.

Fibroblasts are stromal cells that synthesize extracellular matrix, respond to mechanical and inflammatory cues, and shape tissue repair programs through paracrine signaling and matrix remodeling.

Research relevance and current trends

  • Primary fibroblasts are used to study donor-specific matrix production and activation-state changes in vitro.
  • Researchers increasingly examine fibroblast heterogeneity across tissues and disease states.
  • Co-culture systems are commonly used to evaluate stromal effects on epithelial, immune, or tumor-associated phenotypes.

Common research applications

  • Measure matrix production, activation markers, and cytokine responses under defined stimuli.
  • Study wound-healing, fibrosis-associated, or stromal support phenotypes in tissue-relevant contexts.
  • Use in co-culture assays to evaluate paracrine effects on neighboring cell populations.

Product-specific data supplied for this listing

  • Growth Conditions: For optimal cell culture, we recommend using PriCoat™ T25 Flasks (G299) or coating your preferred vessels with Applied Cell Extracellular Matrix (G422) . PriGrow X Series Medium (TM5409) + 1% Penicillin/Streptomycin Solution (G255), 37.0°C, 5% CO₂.
  • Warranty: abm warrants that cell lines shall be viable upon initiation of culture for a period of thirty (30) days after shipment and that they shall meet the specifications on the applicable abm Material Product Information sheet, certificate of analysis, and/or catalog description. Such thirty (30) day period is referred to herein as the "Warranty Period”.
  • Seeding Density (cells/cm²): 7,000 - 10,000

Notes for experimental interpretation

  • Primary cells can show donor-dependent and passage-dependent variation, so morphology, growth rate, and marker expression should be interpreted in the context of the specific lot and culture history.
  • Attachment matrix, medium formulation, and gas conditions can materially influence phenotype maintenance and experimental readouts; use the recommended culture system where possible.
  • Cryopreserved recovery conditions matter for viability and downstream behavior. We recommend using serum-free CryoGuard™ Freezing Media (TM078).

🧊 Thawing Protocol
  1. Thaw cells quickly in a 37°C water bath while agitating gently (maximum 2 minutes). The vial cap should be kept above the water level to minimize the risk of contamination.
  2. Decontaminate the vial by spraying and wiping the exterior of the vial with 70% ethanol. From this point onwards, all operations should be strictly carried out inside a biological safety cabinet using aseptic conditions.
  3. Transfer the cell suspension into a T75 containg 14ml of pre-warmed, complete growth media. Do not centrifuge cells.
  4. Incubate the cells at the recommended conditions.
  5. Allow cells to grow undisturbed for at least 24 hours. Then change the complete growth medium, to remove traces of DMSO.
  6. Change growth medium to fresh, complete growth medium every other day until cells reach 60% confluency. Then, double the complete growth medium volume for cells >60% confluent or weekend feeding.
🔬 Subculture Protocol
Volumes given below are for a T75 flask; proportionally increase or decrease the volume as required per culture vessel size. Subculture cells once the culture vessel is 80% confluent. Trypsinize cells at room temperature; do not warm to 37.0°C.
  1. Aspirate the culture media, wash the adherent monolater with HBSS, and add 10ml of 0.25% Trypsin-EDTA to the culture vessel. Swirl the flask to ensure all cells are covered and then immediately remove 9ml of the Trypsin-EDTA.
  2. Observe the cells under a microscope to confirm detachment (typically within 3-5 minutes). Cells that are difficult to detach can be incubated at room temperature for some additional time. Detach cells by gently tapping the sides of the culture vessel, one cells have rounded.
  3. Neutralize Trypsin-EDTA by adding an equal volume of the complete growth media supplemented with additional 10% FBS into the culture vessel.
  4. Transfer the culture suspension into a sterile centrifuge tube, and centrifuge at 220xg for 5 minutes.
  5. Aspirate the supernatant, and re-suspend the pellet with pre-warmed fresh complete growth media. Add appropriate aliquots of the cell suspension to new culture vessels, as desired.
  6. Incubate the cells at the recommended conditions.
