Assay Principle
This sandwich ELISA quantifies human retinol-binding protein 4 (gene RBP4) in cell culture supernatants, cell lysates, serum, plasma(heparin, EDTA) and urine. Strips are pre-coated with a capture antibody. After the sample is added, a biotinylated detection antibody binds the captured protein, and an avidin–biotin–peroxidase complex supplies the enzyme. TMB develops color in proportion to the bound target; the reaction is stopped and read at 450 nm, and concentrations are read from the standard curve.
Retinol-binding protein 4 (RBP4) is the main plasma carrier of vitamin A. Produced mostly by the liver, it transports retinol in blood, often in complex with transthyretin, and delivers it to tissues through the receptor STRA6. RBP4 is also released by fat tissue and is measured in studies of vitamin A status, insulin resistance and metabolic disease.

Sample dilution: start at 1:20000. In the supplier’s tests, serum and plasma samples measured about 28 µg/mL.
Example standard curve (OD450, ng/mL): 0 = 0.044; 0.312 = 0.129; 0.62 = 0.218; 1.25 = 0.341; 2.5 = 0.569; 5 = 0.937; 10 = 1.445; 20 = 2.112. Run a new standard curve on every plate.
Storage & Handling
Store at 4 °C for up to 6 months or −20 °C for up to 12 months. Avoid repeated freeze–thaw cycles. Ships on gel ice and tolerates up to 3 days at room temperature; freeze on arrival.
