Assay Principle

This sandwich ELISA quantifies human thrombomodulin (gene THBD) in cell culture supernatants, serum, plasma (heparin, EDTA, citrate) and urine. Strips are pre-coated with a capture antibody. After the sample is added, a biotinylated detection antibody binds the captured protein, and an avidin–biotin–peroxidase complex supplies the enzyme. TMB develops color in proportion to the bound target; the reaction is stopped and read at 450 nm, and concentrations are read from the standard curve.

Thrombomodulin (THBD, CD141) is an endothelial surface receptor that binds thrombin and switches it from a procoagulant enzyme to an activator of protein C. The resulting activated protein C pathway limits clotting, and thrombomodulin also affects fibrinolysis, inflammation and angiogenesis. It is studied in vascular biology and thrombosis, and CD141 marks a subset of human dendritic cells.

Example standard curve (OD450, pg/mL): 0 = 0.029; 62.5 = 0.092; 125 = 0.148; 250 = 0.267; 500 = 0.477; 1000 = 0.91; 2000 = 1.518; 4000 = 2.094. Run a new standard curve on every plate.

Storage & Handling

Store at 4 °C for up to 6 months or −20 °C for up to 12 months. Avoid repeated freeze–thaw cycles. Ships on gel ice and tolerates up to 3 days at room temperature; freeze on arrival.
