The HVEM/NFκB Reporter Lentivirus constitutively expresses the human HVEM (TNFRSF14) checkpoint receptor and couples its signaling to an NF-κB-driven dual secreted Gaussia luciferase and fluorescent reporter. Background

HVEM (herpesvirus entry mediator, TNFRSF14) is a tumor necrosis factor receptor superfamily member with a complex role in immune regulation. It serves as a molecular switch that can deliver either stimulatory or inhibitory signals depending on its binding partner: engagement of TNF-family ligands such as LIGHT promotes NF-κB-driven activation, whereas interaction with the immunoglobulin-family receptors BTLA and CD160 transmits inhibitory signals that suppress lymphocyte activation. Through this bidirectional signaling, HVEM helps balance immune activation and tolerance. Its function as a checkpoint and co-stimulatory hub makes HVEM a target of interest in immunotherapy, and NF-κB reporters provide a quantitative readout of HVEM pathway engagement.

Product Description & Applications

The HVEM/NF-κB Reporter Lentivirus is an immunotherapy reporter system for studying HVEM as an immune checkpoint molecule. The construct constitutively expresses the human HVEM receptor; engagement of inhibitory ligands such as BTLA suppresses NF-κB signaling and reduces reporter activity, while agents that block this inhibition restore NF-κB-driven reporter expression. Dual reporters, secreted Gaussia luciferase and a fluorescent protein (GFP or RFP), allow real-time monitoring by luminometry and fluorescence.

Applications include testing immune checkpoint inhibitors that target HVEM signaling and studying lymphocyte regulation. Supplied as high-titer, VSV-G-pseudotyped lentiviral particles purified by PEG precipitation and sucrose gradient centrifugation, the product is optimized for difficult-to-transfect primary and thawed cells.

About This Product

This 2-vial immunotherapy reporter system consists of a Vial 1 Receptor Lentivirus encoding human HVEM under a constitutive promoter with antibiotic selection, and a Vial 2 Reporter Lentivirus encoding tandem NFAT (or NF-κB) response elements driving a dual reporter (GFP, GFP-P2A-GLuc, GLuc, GLuc-P2A-GFP, GLuc-P2A-RFP, RFP, RFP-P2A-GLuc). Sequential transduction and selection generates a dual-stable effector cell line that responds quantitatively to receptor stimulation with a ratiometric fluorescent + bioluminescent readout.

Secreted Gaussia luciferase (where included) accumulates in conditioned media, enabling kinetic sampling without cell lysis. The combined fluorescent and luminescent outputs allow parallel microscopy-based visualization and plate-reader luminometry from the same cell population — providing assay redundancy and flexibility for potency testing formats compliant with regulatory expectations for cell-based functional assays.
