Assay Principle
This microplate immunoassay quantifies Ianalumab by colorimetric detection: samples and kit standards are processed in parallel, and absorbance values are converted to concentrations through the per-plate standard curve.
Background
Ianalumab (VAY736) is an anti-B-cell activating factor (BAFF) receptor fully human IgG1 monoclonal antibody, engineered for direct ADCC-mediated B-cell depletion, thus providing a dual mode of action and targeted approach to treat pSS. The current phase 2b study aimed at establishing a dose-response relationship over a wide range of VAY736 doses, using change from baseline (BL) in the EULAR Sjogren’s Syndrome Disease Activity index (ESSDAI) over 24 Weeks (Wks) as the primary endpoint. Here we report Wk24 efficacy and safety. The study is ongoing with a second blinded treatment period up to Wk52.

Performance Data
Performance is defined by a quantitative range of 0.31-5 μg/mL, an analytical sensitivity of 0.156 μg/mL, typical spike recovery of 80-120%. For best precision, target the central portion of the curve and confirm matrix behavior with spike-recovery and dilution-linearity checks before routine runs.
Target Notes
The target is also referenced in the literature as VAY-736. Curated target information is available under UniProt accession Q96RJ3. The corresponding compound is registered under CAS 1929549-92-7.
Common Research Applications
- Pharmacokinetic profiling of the drug in preclinical and translational research models
- Bioanalytical method development, bridging, and comparability studies
- Stability and formulation studies requiring reproducible concentration readouts
