Compound Overview
ICeD-2 is an inducer of cell death that can kill HIV-1-infected cells, an effect that depends on HIV-1 protease activity. It potently blocks hydrolysis of Gly-Pro-AMC by the dipeptidyl peptidases DPP8 and DPP9, and it strongly stabilizes DPP9 in PBMCs[1]. It is supplied as a white to off-white solid (C20H29N3O, MW 327.46).

In Vitro
ICeD-2 potently blocks hydrolysis of Gly-Pro-AMC by the dipeptidyl peptidases DPP8 and DPP9. ICeD-2 shows exquisite selectivity across this panel of peptidases, with a selectivity window of >3000× fold over both DPP4 and DPP7[1]. In WT THP-1 cells, ICeD-2 treatment results in loss of GFP-positive cells, whereas ICeD-2 has little to no effect on GFP-positive cell numbers when CARD8 (caspase recruitment domain family member 8), CASP-1 (caspase-1), or GSDMD (gasdermin-D) is absent[1].
